报告摘要:

Drug resistance from highly heterogeneous cancer cells represents a major challenge in clinical oncology. Early detection, targeted drug delivery, and precision surgical removal are essential components of successful cancer treatment. The development of multifunctional nanoparticles holds great promises for the effective treatment of drug-resistant cancer. We have developed an integrated image-therapy protocol that takes advantage of the multi-functionality of magnetic iron oxide nanoparticles (IONPs) and advanced methods for targeted imaging and drug delivery, monitoring of therapeutic responses, and optical image-guided surgery. Theranostic IONPs developed by our group are targeted to cellular receptors highly expressed in human cancers. These receptor-targeted IONPs have the capacity of carrying single or multiple therapeutic agents, delivering near infrared (NIR) optical and spectroscopic images, and enabling MRI contrast enhancement. Experiments have demonstrated specificity and sensitivity of optical and MR imaging of tumors using these targeted contrast agents in human pancreatic, breast, and ovarian cancer models in mice.  Further, our results have shown the efficacy of targeted therapy and MRI-monitoring of drug delivery and treatment response after systemic delivery of targeted IONP drug carriers in animal tumor models. Using a hand-held optical imaging device, we have successfully detected and removed small tumor lesions in a surgical cavity to prevent local recurrence and distant metastasis. Our current efforts focus on pre-clinical studies aimed at translating the image-guided therapy into clinical applications, improvement of nanoparticle drug delivery by developing nanoparticles that are able to break physical drug delivery barrier and mechanisms of interactions of receptor-targeted nanoparticles with different cell populations in human tumor tissues.   

 

报告人简介:

Dr. Lily Yang is Professor and Nancy Panoz Chair of Surgery in Cancer Research at Emory University School of Medicine. She received her medical training at West China University of Medical Sciences and the Chinese Academy of Preventive Medicine. She received her PhD degree in Molecular and Cellular Biology from Brown University in 1993.  Dr. Yang’s research focuses on the development of novel nanotechnologies and imaging methods for early cancer detection, targeted drug delivery, non-invasive assessment of therapeutic response, and image-guided surgery.
